Why Meat Gets Tough — and How Marinades Fix It

Meat toughness comes down to two things: connective tissue (primarily collagen) and muscle fiber tension. When an animal uses a muscle heavily — think beef chuck, pork shoulder, or chicken thighs — the fibers are dense and the collagen is abundant. A marinade can’t reverse that entirely, but it can do two genuinely useful things: break down surface proteins to improve texture, and carry flavor deep enough to matter.

The two main tools inside a marinade are acids and enzymes. They work differently, affect different parts of the meat, and have different failure modes. Getting them right — and knowing when one beats the other — is the core skill here.

Acidic Marinades: How They Work and Where They Fail

Acidic ingredients — vinegar, citrus juice, buttermilk, yogurt, wine, beer — denature surface proteins by disrupting their hydrogen bonds. That initial change makes the outer layer of meat feel more tender and helps carry flavoring compounds inward.

But acidity is a double-edged tool. Leave protein in a strongly acidic marinade too long and the same denaturation that softens it starts to firm it up, squeezing moisture out and producing a mushy, almost cooked-looking exterior that turns leathery in the pan.   • Buttermilk and plain yogurt: The mild lactic acid (pH roughly 4.5) works slowly and gently, making these excellent for overnight marinades. Buttermilk-marinated fried chicken can safely rest 24 hours without the meat turning mushy.
  • Wine and beer: Alcohol plus moderate acid. Lower acid concentration means a softer effect; the compounds in wine also contribute subtle tannins that interact with surface proteins.

These are starting points — they depend on acid strength and whether salt and oil are also present. Salt draws moisture inward (effectively brining), which partly counteracts the drying effect of acid. Enzymatic Marinades: The Science of Natural Meat Tenderizers

Enzymes are the more targeted tool. Unlike acids, which denature proteins somewhat indiscriminately, specific plant enzymes cut actual peptide bonds — they literally snip apart muscle fiber proteins (myosin and actin) and, to varying degrees, collagen. The result, when timed correctly, is meat that’s structurally looser without the mushy crust that acid over-marinating creates.

Bromelain (Pineapple)

Found in fresh pineapple juice and the core. Bromelain is powerful — aggressive enough that if you marinate a chicken breast in pure pineapple juice for more than about 30–45 minutes, you can end up with an unpleasantly mushy texture. Canned or cooked pineapple doesn’t work: heat denatures bromelain, which is why a pineapple upside-down cake doesn’t turn your gelatin dessert into liquid.

Best use: diluted in a marinade with oil, soy sauce, and aromatics. Give thick pork shoulder or beef flank 1–3 hours maximum. The result, when calibrated, is remarkably supple without being pulpy.

Papain (Papaya)

Derived from raw papaya (concentrated in the skin and unripe flesh). This is the enzyme behind commercial “meat tenderizer” powder. Papain is slightly less aggressive than bromelain and has a broader temperature range of activity — it remains active even at relatively high cooking temperatures, which means it continues working briefly at the start of the cook. That said, the practical benefit mostly happens before cooking, in the marinade phase.

Using whole papaya: puréeing a few tablespoons of raw, unripe papaya into a marinade paste is a traditional technique across South and Southeast Asian cooking. Ripe papaya contains less papain, so less-ripe fruit is more effective.

Actinidin (Kiwi)

Kiwi contains actinidin, a cysteine protease that tenderizes primarily by attacking myosin. It’s gentler than bromelain and leaves a pleasant, mild flavor. Marinating steak in puréed kiwi for 30–60 minutes is a technique used in some Korean BBQ preparations. Don’t go much longer — the texture gets grainy.

Zingibain (Fresh Ginger)

Fresh ginger contains a protease called zingibain. The effect is milder than the three above, making ginger an ideal supporting tenderizer that works in longer marinades without risk of over-tenderizing. This is why many traditional Asian braises start with a ginger-heavy paste: the enzyme has time to work before cooking, while the flavor contribution is obvious.

Note: dried ginger powder has little to no enzymatic effect — the drying process inactivates the enzyme.

Ficin (Fig)

Fresh fig juice or puréed figs contain ficin, another cysteine protease. Less commonly used in home cooking but sometimes found in restaurant preparations, particularly for lamb. Moderately aggressive.

Combining Acids and Enzymes Strategically

You don’t have to choose one or the other — in fact, the most effective marinades often use both. A marinade with yogurt (mild acid) plus fresh ginger (enzyme) plus a little soy sauce (salt) is genuinely more effective than any single component alone because:

  1. The lactic acid gently opens the surface proteins and begins carrying flavor in.
  2. The enzyme works on the structural proteins without the rapid over-denaturing that strong acid causes.
  3. Salt draws liquid inward (osmosis), which distributes both flavoring and enzyme more deeply.
  4. Oil, when included, slows the marinade’s surface action enough to prevent exterior mush while the interior catches up.

A Base Formula That Actually Works

For about 500–700 g of meat, a flexible starting point:

  • Mild acid: 3–4 tablespoons yogurt or 2 tablespoons buttermilk or 60 ml wine
  • Enzyme source: 2–3 tablespoons freshly grated ginger or 2 tablespoons puréed fresh papaya
  • Salt: 1 teaspoon (or 2 tablespoons soy sauce)
  • Oil: 2 tablespoons neutral or olive oil
  • Aromatics: garlic, herbs, spices to taste

Marinate at refrigerator temperature. If you’re using strong enzymes (bromelain, papain), keep it to 1–2 hours. With milder enzymes and lactic acid, 6–12 hours is comfortable.

Surface Area, Temperature, and the Salt Question

A few variables that determine whether your marinade actually does what you intend:

Surface area matters more than volume of marinade. Marinades don’t penetrate deeply — typically only a few millimeters in a reasonable time frame, regardless of how much liquid you use. Scoring the meat or butterflying it dramatically increases the surface exposed, which is why Korean galbi (cross-cut short ribs) or scored lamb leg marinates so effectively.

Always marinate in the refrigerator. Room-temperature marinating speeds enzyme and acid action but also accelerates bacterial growth. The time gains aren’t worth the food safety trade-off, especially for poultry and pork.

Salt is not optional. A salt-free marinade mostly stays on the surface. Salt shifts osmotic pressure and actively pulls the marinade inward, while also seasoning muscle fibers rather than just coating them. Never reuse marinade. Raw meat transfers pathogens into the liquid. If you want to use marinade as a sauce, reserve a portion before the meat goes in, or bring the used liquid to a rolling boil before serving.

Frequently Asked Questions

Can I use lemon juice as the only tenderizer for a tough steak? For very tough cuts like a flank or skirt steak, lemon juice alone is marginal. It softens the surface but doesn’t penetrate deeply enough to change the texture of the interior. Combine it with a scoring technique, a salt brine (even briefly), and a genuine enzyme source if you want a noticeable structural change — or accept that some cuts need slow braising, not a marinade.

Does marinating in the freezer work? Freezing halts both enzymatic and acid-based tenderizing almost entirely. The popular idea of marinating in the freezer (while the meat defrosts) does produce some light effect during the thawing phase, but you shouldn’t rely on it. Proper marinating needs to happen at refrigerator temperature after the meat is fully thawed.

Why did my pineapple-marinated chicken turn to mush? Bromelain is extremely potent and very fast. If the marinade had a high concentration of fresh pineapple juice and the meat sat in it for several hours — or if it was warm — the enzyme digested the structural proteins past the point of no return. To fix this: dilute the pineapple heavily (roughly 1 part pineapple to 3–4 parts other liquid), keep marinating time to 30–60 minutes, and marinate cold.

Does cooking temperature affect how well an enzymatic marinade worked? Most plant proteases (bromelain, papain, actinidin) are denatured by heat — typically above around 60–70 °C — so they stop working once cooking begins. The tender texture you get is the result of what happened during the marinade phase, not during cooking. The exception is papain, which stays active at slightly higher temperatures and briefly continues working as the meat heats up, which is partly why papain-based commercial tenderizers are sometimes sprinkled on and left at room temperature just before grilling.

Do store-bought meat tenderizer powders work as well as fresh fruit? Commercial tenderizer powders are typically papain extracted and dried, sometimes with salt and other additives. They’re effective and consistent, but they deliver enzyme without the flavor contribution that fresh papaya, ginger, or kiwi brings. They can also over-tenderize if applied liberally and left too long, producing that slightly spongy interior texture that’s a tell in cheap steakhouse kitchens. Tenderizing with acids and enzymes works reliably when you respect the mechanisms behind each ingredient. Acids work on the surface and are forgiving in lower concentrations (buttermilk, yogurt, diluted wine); at high concentrations they damage texture quickly. Enzymes like bromelain are fast and powerful, while zingibain in ginger is slow and gentle — know which you’re using and time accordingly. Combine them with salt and oil for the best effect.
